| 講演者 | Dr. Wei-Li Hsu <要旨> The orf virus (ORFV) is a promising novel viral vector candidate for cancer therapy. Initially, we found that the wildtype ORFV effectively killed nasopharyngeal carcinoma (NPC). To enhance the safety profile of viral vectors for medical applications, two attenuated ORFVs, each deficient in one of the virulence factors, VEGF or CBP, were generated. Their oncolytic effects were subsequently evaluated. Despite this attenuation in animals, efficient infection of the two recombinant ORFVs causes the death of NPC cells. ORFV infection induces pyroptosis in NPC cells, accompanied by significant cell detachment and decreased FAK and AKT activation at the early stage of infection. The innate immune response, particularly, chemotaxis of NK cells,is enhanced by ORFV infection, which lead to increase in the cytotoxicity of NPC cells. Moreover, wild-type and CBP-deleted ORFVs infection significantly inhibited tumor growth in a xenograft mouse model. Due to its potential for anti-tumor activity, the attenuated CBP-deletion ORFV holds promise as a safer viral vector for medical applications. |
