| 講演タイトル | "Systematic analysis of regulators for lipid composition" ＜要旨＞ Beyond making boundaries, membrane lipids have many important functions that are acquired thanks to their extreme structural diversity. Membrane lipid composition differs at various scales, from organisms to organelle subdomains. It is critical to understand better how lipid compositions are regulated through lipid metabolism and exchange between organelles. We used a highly efficient CRISPR-Cas9 strategy to mutate systematically enzymes regulating lipid compositions. This allowed us to have a comprehensive view for the regulation of lipid tail composition between classes. In addition, we found that the loss of individual enzymes can be largely compensated by the action of other enzymes, showing the robustness of lipid synthesis. Moreover, the analysis of lipid changes in various mutants revealed that peroxisomes are capable of feeding de novo lipid synthesis for the whole cell. On the other hand, mitochondria are more “selfish”, having lipid synthetic machineries more suited for local phospholipid production, while acquiring other lipids from the endoplasmic reticulum. Thus, our systematic analysis revealed the redundant and non-redundant aspects of lipid metabolism, contributing to a better understanding how cell type-dependent differences in lipid compositions emerge. |
